Comment: Tirzepatide (LY3298176) is a clinical-stage dual GIP/GLP-1 agonist (‘incretin) that was developed to help manage type 2 diabetes mellitus (T2DM) [1,4] and as a novel anti-obesity medication. Structurally, a 39 amino acid linear peptide conjugated to a C20 fatty diacid moiety by a linker connected to the lysine20 residue. The dual agonist approach synergises the insulin and glucagonostatic responses compared to a GLP-1 agonist alone. It was the first dual-targeting single agent to be approved for T2DM management.
